Update IFU: Citric Acid Test, HOS Test, Fructose Test and LeucoScreen (14-JUN-2017)

The instructions for use for the Citric Acid Test, HOS Test, Fructose Test and LeucoScreen have been updated. They now include a barcode which can be scanned by the app “REA Pharmascan”. This will lead you to the product demonstration video. This video can also be found per product page on our website (see website links below).


Citric Acid Test

To avoid the formation of precipitation during the analysis, we advise to use the semen plasma instead of the whole semen sample (see specimen type section). The centrifuge should be set up at 2500g for 20 minutes after addition of Reagent 2. Further, a detailed instruction on how to pipette the supernatans (25µL) and the assay performance parameters are indicated in the IFU of the Citric Acid Test.

HOS Test

More attention is paid to the percentage of tail abnormalities before exposure to the hypo-osmotic medium (=step 1). This percentage should be subtracted from the calculated result with the HOS Test (see section Result).

Fructose Test

We have added in the method that we recommend to use a fitting heat block in step 11, if available in the lab. The wavelength range measured with the plate reader / photometer is extended from a 470-492nm to a 450-492nm filter. Further, the specimen type and assay performance parameters are mentioned on the IFU.

The interpretation will now also be easier if you download the excel calculation sheet for the Fructose Test on our website.

LeucoScreen

The protocol should be performed under a fume hood, as Reagent 1 is poisonous. More details are given on the interpretation of the cells, the specimen type and the number of test days.

The instructions for use for the SpermMar Test IgA, SpermMar Test IgG (and controls), EpiScreen Plus and Spermac Stain have been updated. They now include a barcode which can be scanned by the app “REA Pharmascan”. This will lead you to the movie of the product. This movie can also be found per product page on our website (see website links below).

The test protocol of the SpermMar Test IgA and SpermMar Test IgG (Positive and Negative controls) has not changed however.

Step 3:“Let the slide dry by placing it on a warm plate at 37°C for 15 minutes.”
Step 5: “Stain 2 minutes in stain A” (instead of 1 minute)

SpermFreeze SSP and Fructose Test IFU update + calculation sheet Fruct (17-NOV-2016)
The instructions for use for SpermFreeze SSP and the Fructose Test have been updated. For the SpermFreeze SSP the test protocol has not changed however. The instructions for use are translated in 5 languages and the instructions for use bear the CE mark now.

The instructions for use of the Fructose Test are updated with the remark that steps with Reagent 3 should be performed under a fume hood:
step 9: "Remark: perform step 10-13 under a fume hood as Reagent 3 is toxic by inhalation (see warnings and precautions)."
Warnings and precautions: "Reagent 3 (Indole in methanol): ...Always work under a fume hood when using this reagent."

Update LeucoScreen instructions for use (21-APR-2016)
The instructions for use for the LeucoScreen have been updated. The test protocol has not changed however.

Based on customer feedback and observations of the R&D department the peroxidase-positive white blood cells can be better distinguished from other round cells when the concentration H2O2 is lowered. The amount of air bubbles in positive samples is also reduced with a lower concentration H2O2. Therefore the H2O2 concentration in Reagent 2 is lowered from 30% to 3%.

Update GAIN instructions for use (19-FEB-2016)
The instructions for use for GAIN medium have been updated. The intended use and test protocol has changed !

Intended use:
GAIN medium consists of 2 types of ready to use cell culture media for use with human embryos and gametes. The media can be used for fertilization, zygotes and embryos culture through the cleavage stages and up to the expanded blastocyst stage and for embryo transfer.

  • GAIN medium Early Stage (GAIN ES) can be used for the handling of oocytes (in preparation of, or during IVF/ICSI) and for cell culture from fertilization up to 4-cell stage, for the processing of semen and for embryo transfer.

  • GAIN medium Blastocyst (GAIN BL) is designed for embryo culture from 4-cell up to expanded blastocyst stage and for embryo transfer. Alternatively, the medium can be used immediately after fertilization, up till expanded blastocyst stage.
  • Update HOS test instructions for use (16-DEC-2015)
    The instructions for use for the HOS test have been updated. The test protocol has not changed however.
